Description

With Mars expeditions slated for the 2030's, tissue-specific surrogate end-points are required to accelerate the investigation of the long-term health effects of space radiation including cancer and degenerative diseases. Additionally, state-of-the art biological models including but not limited to 3-D culture, organoids, 3-D bioprinting, tissue chips, humanized animals, and systems biology approaches need to be identified and/or developed to support assessment or radiation-associated cancer risk and shared biology with degenerative diseases. Understanding the molecular, cellular, and tissue responses to space radiation exposure is necessary to identify surrogate end-points for pre-malignancy and degenerative diseases and use systems biology approaches to extrapolate to human risk as well as identify and evaluate countermeasure efficacy. The overall goal will be to identify tissues-specific surrogate end-points for accelerated assessment of radiation-associated carcinogenesis and degenerative diseases and future countermeasure development and testing.

Approach
Data mining of both animal and human clinical data for identification of potential tissue-specific surrogate end-points. Tech watches and expert/government inter-agency knowledge for identification of state-of-the art biological models. Ground-based research to assess space radiation-induced cancer initiation, promotion, and progression (and shared biology with other disease states), and identify and validate both biological model systems and tissues-specific surrogate end-points.

  • Identification of biological model systems to evaluate radiation-associated outcomes related to carcinogenesis and degenerative diseases.
  • Identification of sensitive and specific surrogate end-points that are validated in appropriate animal models and human cohorts.
