Aims:
We propose that endogenous neural stem and precursor cells constitute a pool of redox sensitive cells that are radioresponsive targets for low dose radiation effects. We further contend that low dose irradiation is a circumstance that can precipitate age-related disorders in the CNS through the induction of oxidative stress. We hypothesize that the activation of redox sensitive signaling can trigger radioadaptive changes in these cells that can be either harmful or beneficial to the overall cognitive health of an organism. To test these ideas, we will irradiate and analyze neural precursor cells grown in culture as well-defined 3-dimensional neurospheres (aims 1, 2) and we will irradiate mice to assess the impact of low dose exposures on their ability undergo neurogenesis (aims 3, 4).
