Aims:
Specific Aim 1: Characterize oxidative lung damage from repeated Radiation/hyperoxia
exposure in a novel mouse model of space-flight related challenge.

Mouse cohorts will be subjected to repeated rounds of sublethal (0.25Gy) total body
radiation, 8 hours of hyperoxia (100% O2) or a combination of both, 3 times weekly for
4 weeks and evaluated at 1 month (acute) and 2,3 and 4 months (Chronic) later for
oxidative damage, fibrosis and inflammation of lung tissues. Brain and blood tissues will
also be evaluated to determine systemic toxicity.

Specific Aim 2: Evaluate usefulness of dietary FS and FLC as countermeasure to oxidative
lung injury in a mouse model of radiation/hyperoxia relevant to space flight challenges.

Mouse cohorts will be placed on a flaxseed (FS) or flaxseed lignan component (FLC)
diets and subjected to the conditions described in Aim1. Lung tissues as well as Brain,
and blood will be collected after 1 month (acute) and compared to that collected after 2,3,
4 months (Chronic) with respect to oxidative damage, inflammation, fibrosis.
